    Summary of Direct Charitable Activities
    List the foundation’s four largest direct charitable activities during the tax year. Include relevant statistical information such as the number of organizations and other beneficiaries served, conferences convened, research papers produced, etc. Expenses
    1 While the focus of the Jain Foundation is LGMD2B, the resulting scientific advances and collaboration have a broader impact on human health through their relevance to other muscle disorders, degenerative diseases of tissues other than muscle, and normal aging. The Foundation is hands on in its approach to funding. We closely monitor progress (every quarter) of every project we fund and keep track of results and agreed upon timelines through detail oriented conference calls with researchers and site visits. Our goal continues to be to fund all possible approaches to a cure and have the patients diagnosed for when we reach a cure so they are able to avail it. Contract Research Organizations: In 2012 we initiated several projects at Contract Research Organizations. There are many drugs in development (many even FDA approved) for other diseases that may be useful in LGMD2B. To evaluate their benefit in LGMD2B we need specific assays. The process of developing assays is tedious, time consum 488,206
    2 Patient Recruitment and Diagnosis: LGMD2B is very rare. An important goal of the Foundation is to identify LGMD2B patients worldwide. The Foundation maintains a patient registry and facilitates by providing funding for patients' diagnosis. The Jain Foundation team reviews patient records to identify patients that have a high probability of having dysferlinopathy before doing expensive protein or genetic analysis covered by the Foundation. More often than not, however, patients records or their physicians do not have the information needed for us to make a well-informed decision. In an attempt to identify patients rapidly and to save money, the Foundation developed an easy-to-use online diagnostic tool. The tool is based on a Bayesian algorithm and is grounded on published clinical reports. The tool discriminates between 25 types LGMD. The tool was launched in May 2012 and has been very well received by the medical community all over the world. This tool has increased our rate of findin 115,454
    3 Clinical Outcome Study: The process of becoming trial ready was started in 2012. Clinical trials frequently fail because testable outcomes are not well defined and are not sensitive enough to measure a small benefit from the treatment or drug being delivered. To find measurable outcomes for dysferlinopathy we launched a Clinical Outcome Study (COS) running out of 14 centers (in the US, Europe, Japan and Australia). COS will recruit 150 patients. Patients will visit their respective centers 6 times over 3 years for medical, physiotherapy and MRI/MRS assessments at each visit. We are funding, leading and managing this study. COS will help define the best tests for measuring disease progression. We will also collect and bank biological samples for use in research and identify and register as many dysferlin deficient patients as possible worldwide. Finally COS will get all 14 sites ready for clinical trials. In other words, COS is a dress rehearsal for future clinical trials. 82,624
    4 Tools for Researchers: The Jain Foundation continues to actively identify and fund the development of research tools and resources that were needed to advance research in the dysferlin field in 2012. The Foundation is also committed to developing and providing frequently required reagents, the unavailability of which slows down research. In addition we generated and maintained new mouse models at a custom breeding colony at Jackson Laboratories (Maine, US).
