Cancer Computational biology superstar Sanju Sinha joins Sanford Burnham Prebys Sanju Sinha, Ph.D., has joined Sanford Burnham Prebys as an assistant professor in the Cancer Molecular Therapeutics Program to continue his research on cancer development and drug discovery. Cells need more than an accumulation of mutations to become cancerous-they must be triggered into becoming cancerous by changes in their environment. One of the goals of Sinha's research is to identify these triggers and develop interventions against them. Reviving exhausted T cells to tackle immunotherapy-resistant cancers When the cells of our immune system are under constant stress due to cancer or other chronic diseases, the T cells of the immune system shut down in a process called T-cell exhaustion. Without active T cells, which kill tumor cells, it's impossible for our bodies to fight back against cancer. Researchers at Sanford Burnham Prebys, including Linda Bradley, Ph.D. and Jennifer Hope, Ph.D., described a new approach in Cell Reports that can reduce T-cell exhaustion even in melanoma tumors that are resistant to clinically approved immunotherapies. It can also help T cells from becoming exhausted. Taking a 3D view of the genome may help treat pediatric brain cancers Researchers at Sanford Burnham Prebys, led by Lukas Chavez, Ph.D., are leveraging the latest technology to take a never-before-seen look at ependymoma, one of the deadliest pediatric brain tumors. By visualizing how the genome is organized and arranged within tumor cells, they were able to reveal genes in tumors that may be future targets for therapy. The results appear in Nature Communications. Charles Spruck awarded $1.7M to advance "ancient virus" treatment for prostate cancer With the help of a grant from the U.S. Department of Defense for more than $1.7 million, Charles Spruck, Ph.D., will advance an innovative therapeutic approach for metastatic prostate cancer. Known as viral mimicry, the approach tricks the body into thinking that it has a viral infection, stimulating an immune response that can help the body fight cancer. Sanford Burnham Prebys researchers awarded $2.6M to discover pancreatic cancer drugs Cosimo Commisso, Ph.D., and Susanne Heynen-Genel, Ph.D., earned $2.6 million from the National Cancer Institute to advance a new treatment approach for pancreatic cancer. project will identify potential drugs that can manipulate the pH of cells to stop pancreatic tumors from growing. The promising approach would selectively kill pancreatic cancer cells without affecting surrounding healthy cells. New treatment approach for prostate cancer could stop resistance in its tracks For the first time, researchers have discovered that prostate cancer can be killed by targeting a single enzyme, called PI5P4K. The findings, published recently in Science Advances, could help address the growing threat of treatment resistance in prostate cancer and could also lead to improved treatments for other cancers, such as those affecting the breast, skin and pancreas. Childhood cancer trailblazer Lukas Chavez joins Sanford Burnham Prebys Lukas Chavez, Ph.D., has joined Sanford Burnham Prebys as an assistant professor in the Tumor Initiation and Maintenance Program to continue his research on chromatin and gene regulation in childhood brain cancers. His research focuses on circular DNA fragments called ecDNA that are detached from normal chromosomes. These DNA circles allow cancer cells to produce and transcribe many copies of cancer-causing genes. Liver cancer study encourages caution with certain gene therapies Research led by Randal J. Kaufman, Ph.D., has found that misfolded proteins in liver cells contribute to the development of liver cancer, shedding new light on the mysterious origins of one of the world's deadliest diseases. The findings, published in the journal Molecular Therapy, could also help improve the safety of certain gene therapies for hemophilia. Zhouji Chen, Ph.D., was co-first author on the study. Sanford Burnham Prebys selected for participation in National Cancer Institute Chemical Biology Consortium For the third time, Sanford Burnham Prebys has been selected by the Frederick National Laboratory for Cancer Research, currently operated by Leidos Biomedical Research, Inc., supporting the National Cancer Institute Experimental Therapeutics (NExT) Program as a Center for the Chemical Biology Consortium. Inhibiting key metabolic enzyme shows promise against melanoma Researchers at Sanford Burnham Prebys, led by Ze'ev Ronai, Ph.D., have shown for the first time that inhibiting a key metabolic enzyme selectively kills melanoma cells and stops tumor growth. Published in Nature Cell Biology, these findings could lead to a new class of drugs to selectively treat melanoma, the most severe form of skin cancer. Jianhua Zhao awarded $2.4M to reveal cancer targets through atomic-resolution imaging Jianhua Zhao, Ph.D., has been awarded a unique and competitive grant from the National Institute of General Medical Sciences, part of the National Institutes of Health. The five-year, $2.4 million grant aims to give researchers greater research flexibility to work on fundamental questions in biology. Zhao's research focuses on proteolysis, the process cells use to break down and recycle unused proteins. Proteolysis is essential for normal cell replication because our cells use the by-products of this process to know when it's time to replicate. Because cancer occurs when cellular replication spirals out of control, proteolysis is also an important process for cancer cells. Degenerative Diseases Randal J. Kaufman among the world's most highly cited researchers Sanford Burnham Prebys professor Randal J. Kaufman, Ph.D. was included on Clarivate's 2022 Highly Cited Researchers list, a global ranking of influential researchers based on the number of times their work has been cited in peer-reviewed publication over the last decade. Launched in 2014 by Clarivate, a global research analytics company, the list identifies scientists who have demonstrated exceptional influence in their respective fields. Timothy Huang awarded $2.8M to study well-known gene linked to Alzheimer's disease Timothy Huang, Ph.D., was awarded $2.8 million by the National Institute on Aging (NIA) to continue his work on Alzheimer's disease. One of the defining characteristics of Alzheimer's disease is the accumulation of proteins in the brain called amyloid proteins. Huang and his team will be studying a gene called SORLA, which helps regulate how much amyloid protein is able to accumulate. Development, Aging and Regeneration Program New algorithm can predict diabetic kidney disease Researchers from Sanford Burnham Prebys developed a computational approach to predict whether a person with type 2 diabetes will develop kidney disease, a frequent and dangerous complication of diabetes. Yu Xin (Will) Wang joins Sanford Burnham Prebys to advance regenerative medicine Molecular biologist Yu Xin (Will) Wang, Ph.D., has joined Sanford Burnham Prebys as an assistant professor in the Development, Aging, and Regeneration Program to uncover the ways muscle, nerve and immune cells work together to overcome disease. Wang studies the cell-cell interactions that mediate the function of tissue-specific stem cells during regeneration and disease. When these cell-to-cell interactions go awry, it can result in debilitating neuromuscular diseases, such as muscular dystrophy and myasthenia gravis. Research unlocks the circuitry of diabetes Research led by Pamela Itkin-Ansari, Ph.D., and Randal Kaufman Ph.D., has mapped out a network of biochemical interactions that help special cells in the pancreas called islet cells create insulin, shedding light on the origins of diabetes and revealing new targets for future treatments. The study, recently published in the journal JCEM, found that inflammation disrupts these interactions, triggering a snowball effect of cellular stress that damages pancreatic islets and makes it harder for the body to produce insulin. These findings open up a new world of potential targets to treat and possibly even cure diabetes. Erkki Ruoslahti wins America's top biomedical research award Distinguished Professor Emeritus Erkki Ruoslahti, M.D., Ph.D., has been announced as one of three winners of the Albert Lasker Basic Medical Research Award, the most prestigious American award for biomedical research, for his transformational research leading to the discovery of the cell adhesion receptors now known as integrins. Ruoslahti is the first Institute researcher to ever win the Lasker Award, known as "America's Nobel." Peter Adams and Bing Ren awarded $10.6M to create atlas of aging cells Sanford Burnham Prebys professor Peter Adams, Ph.D., and Bing Ren, Ph.D., from UC San Diego, were awarded a new grant from the National Institutes of Health to join a multi-institution initiative studying the process of aging. The $10.6 million grant was awarded as part of the Cellular Senescence Network program, which aims to create a comprehensive atlas of how and where aging cells accumulate in our tissues and open new avenues for treating age-related diseases. Human Genetics Incurable liver disease may prove curable Research has shown for the first time that the effects of Alagille syndrome, an incurable genetic disorder that affects the liver, could be reversed with a single drug. Sanford Burnham Prebys announces start of Phase 2 clinical trial of DS-1211 in individuals with pseudoxanthoma elasticum A Phase 2 clinical trial has started of DS-1211 in individuals with pseudoxanthoma elasticum (PXE), a rare multisystem genetic disease that causes calcium deposits in soft tissue resulting in considerable morbidity. DS-1211 is a potential first-in-class small molecule developed through a research collaboration between Daiichi Sankyo and Sanford Burnham Prebys. The foundational science for DS-1211 was conducted by Jose Luis Millan, Ph.D. The Conrad Prebys Center for Chemical Genomics played a key role in the development of DS-1211 by leading the drug discovery effort. Immunity and Pathogenesis Pumping the brakes on autoimmune disease In a study published in Structure, researchers at Sanford Burnham Prebys have revealed the structure and function of a drug called LY3361237, which can reduce the harmful activity of the immune system to help treat autoimmune diseases. Their work laid the foundation for a new treatment that's currently in a Phase 2 clinical trial for lupus, an autoimmune disease affecting multiple organs in the body.
